Genom denna teknik s\u00e5 kan man visualisera och f\u00e5 information om molekyl\u00e4ra interaktioner p\u00e5 nanoniv\u00e5.<\/p>\n<p>Nikon har utvecklat N-STORM med ny optik som skapar effektivare illuminering som \u00e4r optimerad f\u00f6r CMOS kameror. Det ger 10 g\u00e5nger snabbare bildtagning j\u00e4mf\u00f6rt med tidigare system och det m\u00f6jligt att skapa STORM bilder ocks\u00e5 av levande celler. Ut\u00f6ver den f\u00f6rb\u00e4ttrade bildtagningshastigheten s\u00e5 finns nu ocks\u00e5 m\u00f6jlighet att ut\u00f6ka det avbildade synf\u00e4ltet till 512 x 512 pixlar. N-STORM tillsammans med det ultra-stabila inverterade mikroskopet Ti2-E, och verktygen i mjukvaran NIS-Elements skapar ett m\u00e5ngsidigt mikroskopsystem som ger anv\u00e4ndaren en o\u00f6vertr\u00e4ffad flexibilitet f\u00f6r m\u00e5nga olika applikationer.<\/p>\n<h3>Tiofaldig f\u00f6rb\u00e4ttring av lateral uppl\u00f6sning (till 20nm)<\/h3>\n<p>N-STORM anv\u00e4nder lokaliseringsinformation fr\u00e5n tusentals individuella fluorokromer f\u00f6r att skapa fantastiska superresolution bilder. Det \u00e4r m\u00f6jligt att f\u00e5 en optisk uppl\u00f6sning som \u00e4r 10 g\u00e5nger h\u00f6gre \u00e4n konventionella optiska mikroskop.<\/p>\n<h3>Tiofaldig f\u00f6rb\u00e4ttring av axiell uppl\u00f6sning (till 50 nm)<\/h3>\n<p>F\u00f6rutom att f\u00f6rb\u00e4ttra den laterala uppl\u00f6sningen s\u00e5 finns ocks\u00e5 m\u00f6jligheten f\u00e5 en 3D STORM bild med tiofaldig f\u00f6rb\u00e4ttring i axiell uppl\u00f6sning. F\u00f6r att kunna avbilda stora volymer s\u00e5 har N-STORM en 3D-Stack-funktion d\u00e4r flera 3D STORM-bilder i olika Z-positioner kombineras samman i en bild.<\/p>\n<h3>Dynamisk Super Resolution<\/h3>\n<p>Bildtagningshastigheten har f\u00f6rb\u00e4ttrats avsev\u00e4rt, fr\u00e5n minuter till sekunder per bild, tack vare nyutvecklad optik och illuminering som \u00e4r optimerade f\u00f6r sCMOS-kameror. Tack vare denna f\u00f6rb\u00e4ttring \u00e4r det nu m\u00f6jligt att avbilda levande celler med en uppl\u00f6sning som \u00e4r 10 g\u00e5nger st\u00f6rre \u00e4n med konventionella optiska mikroskop.<\/p>\n<h3>Flera f\u00e4rger f\u00f6r studier av proteininteraktioner<\/h3>\n<p>F\u00f6r att studera interaktioner mellan flera proteiner s\u00e5 kan STORM bilder tas med upp till 3 fluorochromer, antingen genom den sekventiella aktivator-reporterpar metoden, eller med kontinuerlig aktivering av fluorochromerna.<\/p>\n<h3>H\u00f6gre densitet av molekyler ger h\u00f6gre uppl\u00f6sning<\/h3>\n<p>De nyutvecklade optiken och den f\u00f6rb\u00e4ttrade excitations kapaciteten g\u00f6r att fler molekyler kan lokaliseras per bild. Detta ger en exaktare lokalisering av molekyler och d\u00e4rmed h\u00f6gre uppl\u00f6sning.<\/p>\n<h3>Korrelativ Superresolution f\u00f6r information om molekylernas<\/h3>\n<p>N-STORM kan kombineras med konfokalmikroskopi, till exempel med Nikon\u2019s A1R HD25 konfokalsystem. Fr\u00e5n en konfokalbild med stort synf\u00e4lt s\u00e5 v\u00e4ljer du ett intressant omr\u00e5de och byter till superresolution med ett klick i mjukvaran.
